The Origin and Stimuli Implicated in the Expression of Nestin Cardiac Myocyte- like Cells in the Ischemic Heart

چکیده
Studies from our lab demonstrated that scar formation and healing was associated with the appearance of nestin (+) cardiac myocyte-like cells predominantly at the peri-infarct region. The focus of the present study was to identify the underlying mechanism(s) (e.g. hypoxia, neurohormones) implicated in their recruitment and their cellular origin. The presence of these cells was detected as early as 1-week post-myocardial infarction (MI) and persisted 9 months after complete coronary artery ligation. Furthermore, nestin (+) cardiac myocyte-like cells were also detected in the infarcted human heart. Hypoxia represents a predominant stimulus following MI, however the exposure of normal rats to a hypoxic environment failed to promote the re-appearance of nestin (+) cardiac myocyte-like cells. By contrast, the infusion of the non-selective β-adrenergic agonist isoproterenol (ISO) in the normal adult Sprague-Dawley rat increased nestin expression in the left ventricle and was associated with the reappearance of nestin (+) cardiac myocyte-like cells. However, the reappearance of nestin (+) cardiac myocyte-like cells may not represent a direct effect but was apparently secondary to cardiac myocyte necrosis mediated by isoproterenol. Lastly, we identified a subpopulation of nestin-immunoreactive cells in the normal rat heart that coexpressed cardiac progenitor cell markers Nkx-2.5 and GATA-4. This subpopulation of nestin/Nkx-2.5/GATA-4 cells may represent the progenitor pool that differentiates to a nestin (+) cardiac myocyte-like cell following an ischemic insult.

The phenotype and potential origin of nestin+ cardiac myocyte-like cells following infarction.
Nestin+ cardiac myocyte-like cells were detected in the peri-infarct/infarct region of the ischemically damaged heart. The present study was undertaken to elucidate the phenotype and potential origin of nestin+ cardiac myocyte-like cells and identify stimuli implicated in their appearance. In the infarcted human and rat heart, nestin+ cardiac myocyte-like cells were morphologically and structur...

متن کاملThe Effect of Eight Weeks of Intermittent Training with Quercetin Encapsulated Nanoparticles on JNK and ERK Apoptosis Regulators in the Rat Model of Myocardial Infarction
Introduction: Restoration of blood flow to the ischemic heart muscle causes infarction and myocyte death. Quercetin can be effective in treating heart disease due to its antioxidant effect, but its use is limited due to its low solubility in water. Quercetin restriction can be removed by embedding it in carrier nanoparticles. The present study was designed to determine the effect of quercetin e...

Objective(s): The beneficial and more potent role of exercise to prevent heart apoptosis in ovariectomized rats has been known. The aim of this study was to examine the effects of swimming training on cardiac expression of Bcl-2, and Mir-133 levels and glycogen changes in the myocyte. Materials and Methods: Forty animals were separated into four groups as control, sham, ovariectomy (OVX) and o...
متن کاملDifferential gene expression by lithium chloride induction of adipose-derived stem cells into neural phenotype cells
Objective(s): Adipose-derived stem cells (ADSCs), with suitable and easy access, are multipotential cells that have the ability for differentiation into other mesodermal and transdifferentiate into neural phenotype cells. In this study, Lithium chloride (LiCl) was used for in vitro transdifferentiation of rat ADSCs into neuron-like cells (NLCs).<stro...
